Genomic engineering provides the ability to manipulate gene expression in a variety of desired cell types, including stem cells. This course will provide an overview of the current major genomic engineering platforms such as CRISPR, TALENs, and ZFNs, but it will cover CRISPR genome editing technology in depth.

The first part of the course will focus on the general principles of genome editing protocols, including design, choice of format, delivery, efficiency, specificity, clonal isolation, genotyping, and validation. The second part of the course will address different applications including genome editing in mice, zebrafish, and iPS cells, disease modeling, generation of reporter lines, and high throughput approaches. We will discuss strategies to make CRISPR gene editing more efficient, flexible, and specific. We will explore recent advances in the CRISPR field including base editors and epigenome editing. We will also examine sequencing and quality control considerations for genome editing projects. Hands-on laboratory exercises will accompany the lecture material to provide practical training in design, assembly, transfection, and detection/evaluation steps of a typical genome editing workflow.

General Principles of Genome Engineering; DNA Repair Mechanisms; CRISPR-mediated Gene Modulation, Epigenome Editing, Targeted Integration, Genome Editing Workflow, Gene Correction in iPSCs; Targeted Mutagenesis in Zebrafish; CRISPR Genome Engineering in ES Cells and Embryos for Mouse Model Generation; Developing Reporter Lines; Sequencing and Quality Control Considerations.

Laboratory Topics:
Introduction to Genome Editing Technologies; CRISPR/Cas9 Design Considerations; Transfection of Cas9/gRNA Ribonucleoproteins (RNPs); Gene Knockout Using Targeted Nucleases; Evaluation of Editing Efficiency, Targeted Integration; Donor DNA Design and Nuclease Compatibility; Detection of Integration Events, Exercises for CRISPR Design; Bioinformatics for Genome Editing Applications; Development of Genetically Modified Cell Lines Using K562 Cells (will allow attendees to run their own experiments and analyze their own results by the end of this workshop); and Best Practices for Use of the CRISPR/Cas9 System for Genome Editing.

Each participant will receive Continuing Education Units (CEUs) based on the number of contact hours. Upon completion of the course each participant will receive a certificate, showing completion of the workshop and CEU credits.  For a 3-day workshop, a participant receives 2.1 CEUs.  For a 4-day workshop, a participant receives 2.8 CEUs.  For a 5-day workshop, a participant receives 3.5 CEUs.
